Description
For the detection of Mycoplasma synoviae infection in chickens and turkeys.


Composition
The antigen is a suspension of killed and coloured Mycoplasma synoviae organisms, strain WVU-1853(A.T.C.C.). The agglutinability has been standardized against a standard serum.

Indications
The antigen is used for the detection of a Mycoplasma synoviae infection in chickens or turkeys. Because it is possible that not all birds of an infected flock react positively, it is strongly recommended to use the plate agglutination test for flock diagnosis and not for the individual identification of infected birds. Mycoplasma synoviae infection can be potentially present in a flock. It is therefore recommended only to consider a flock as negative, when the test has been repeated several times and has given a negative result each time.

Test performance
Materials:
- A well cleaned glassplate, illuminated from below (avoid excessive heat from the light source).
- A small stirring rod.

The test should be carried out between 20° and 25°C. At higher temperatures false positive reactions caused by auto-agglutination may be observed.

A drop of serum of approx. equal size is added to a drop of the well-shaken antigen. The drops are then mixed with the stirring-rod and spread to a circular area of approx. 1.5 cm diameter. By rocking the plate a positive agglutination is stimulated. The reaction should be carried out with fresh serum. The serum should not be frozen as this may lead to aspecific reactions.

Test interpretation
Positive reactions show within 2 minutes and are characterized by blue coloured flocculation. The size of the floccules can vary and is also dependent on the concentration of the antibodies in the serum. A reaction can be considered as negative only after an observation period of two minutes. Flocculation occuring after 2 minutes must be considered as a doubtful reaction.
